You do not need to de-activate first.  As long as you have the .mdb file you will be able to see all of your templates and data.  All you need to do is copy the AsureID.mdb from the source computer and replace the one that is on the destination computer.

Here is the path to the database:

On XP:

C:\documents and settings\all users\application data\HID Global\Asure ID\AsureID.mdb.

On Windows 7 and Vista:

C:\Program Data\HID Global\Asure ID\AsureID.mdb

Please keep in mind that both of these locations are in a hidden system folder.  You will need to change your folder viewing options to be able to locate them.
